Product Specific Information
The eBio500A2 monoclonal antibody reacts with the 25 kD epsilon subunit of the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells and NKT cells. Binding of eBio500A2 to CD3 initiates intracellular biochemical pathways leading to cellular activation and proliferation.
The 500A2 antibody partially cross-blocks 17A2 and 145-2C11 antibodies, indicating recognition of distinct but overlapping epitopes.

Target Information
CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. Activation through TCR involves CD3 gamma, delta, epsilon, and zeta subunits, which are structurally related members of the immunoglobulin superfamily encoded on human chromosome 11.
CD3 components have long cytoplasmic tails that interact with signal transduction molecules via double tyrosine-based motifs. CD3 may play roles in TCR-induced growth arrest, cell survival, and proliferation.
CD3 antigen is expressed on:
- 68–82% of normal peripheral blood lymphocytes
- 65–85% of thymocytes
- Purkinje cells in the cerebellum
Not expressed on B or NK cells. Reduced T lymphocyte percentages may occur in autoimmune diseases. Genes encoding CD3 epsilon, gamma, and delta are located on chromosome 11. CD3 gene defects are associated with CD3 immunodeficiency.
