Target Information
IL-33R, also known as ST2, is a receptor for IL-33 and a member of the IL-1 receptor family. Its extracellular domain consists of three Ig-like domains that interact with co-receptor IL-1RAcP and ligand IL-33. Upon IL-33 binding, the IL-33R complex associates with adaptor proteins MyD88, IRAK1, IRAK4, and TRAF6, activating NF-κB and MAPK pathways. This leads to the release of cytokines such as IL-5, IL-6, IL-8, and IL-13.
Two major isoforms exist:
- ST2L: membrane-bound form
- ST2S: soluble form acting as a decoy receptor
IL-33R is primarily expressed by mast cells, basophils, and eosinophils, and in mice, also by ILC2 and Th2 cells. Its signaling is implicated in immune conditions such as allergies, arthritis, atherosclerosis, and sepsis.
