Target Information

Piezo1 is a pore-forming subunit of a mechanosensitive non-specific cation channel that generates currents characterized by a linear current-voltage relationship sensitive to ruthenium red and gadolinium. Structurally, Piezo1 contains 36 transmembrane domains and functions as a homotetramer.

Piezo1 plays a key role in:

• Epithelial cell adhesion via integrin activation and calpain signaling
• Detection of intraluminal pressure changes and urine flow sensing in the kidney
• Shear-stress sensing for endothelial cell alignment and vascular structure formation

Mutations in the PIEZO1 gene are associated with:

• Dehydrated Hereditary Stomatocytosis (with/without Pseudohyperkalemia or Perinatal Edema)
• Lymphedema
