Target Information
Piezo1 is a pore-forming subunit of a mechanosensitive non-specific cation channel. It generates currents characterized by a linear current-voltage relationship that are sensitive to ruthenium red and gadolinium. Structurally, Piezo1 contains 36 transmembrane domains and functions as a homotetramer.
Piezo1 plays a key role in:
- Epithelial cell adhesion via integrin activation through R-Ras recruitment to the ER.
- Detection of intraluminal pressure changes and urine flow sensing in the kidney.
- Shear-stress sensing that promotes endothelial cell organization and vascular development.
Defects in the PIEZO1 gene are associated with:
- Dehydrated hereditary stomatocytosis (with/without pseudohyperkalemia and/or perinatal edema)
- Lymphedema.
