Target Information

STK3 and STK4 are stress-activated, pro-apoptotic kinases which, following caspase-cleavage, enter the nucleus and induce chromatin condensation followed by DNA fragmentation. These kinases are key components of the Hippo signaling pathway, regulating organ size and tumor suppression by restricting proliferation and promoting apoptosis.

The pathway involves a kinase cascade where STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 with MOB1. This in turn phosphorylates and inactivates YAP1 and WWTR1/TAZ, inhibiting nuclear translocation and regulating genes involved in proliferation, apoptosis, and migration.

STK3/MST2 and STK4/MST1 repress hepatocyte proliferation, prevent activation of liver stem cells, and inhibit tumor formation. They also phosphorylate NKX2-1, NEK2, MOBKL1A, RASSF2, and MOBKL1B, playing roles in centrosome disjunction, RAF1 activation, and ESR1 transcriptional activity modulation.
