Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues and cell lines. CD45 plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases. Multiple isoforms of CD45 exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45RA is an isoform with restricted expression among lymphoid subtypes. CD45 is an essential regulator of T- and B-cell receptor signaling, suppresses JAK kinases to modulate cytokine receptor signaling, and promotes cell survival through integrin-mediated pathways and apoptosis regulation.
