Product Specific Information

Description:
The eBioG4.18 monoclonal antibody recognizes rat CD3 protein, a key component of the T-cell receptor (TCR) complex and a marker of the T-cell lineage. CD3 mediates signal transduction from the TCR to the nucleus, leading to cellular responses such as IL-2 transcription and clonal expansion.

In vitro, immobilized G4.18 antibody activates T cells, while soluble G4.18 inhibits allogeneic mixed-lymphocyte proliferation and cytotoxicity. In vivo, it can induce long-term tolerance to organ allografts.

Applications Reported:
Reported for use in flow cytometry, immunoprecipitation, immunoblotting (WB), and immunohistochemical staining.

Applications Tested:
Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.5 µg/test (100 µL final volume). Cell number should be optimized (10⁵–10⁸ cells/test). Antibody titration is advised for optimal performance.

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by closely linked genes on human chromosome 11.

CD3 plays a role in TCR-induced growth arrest, cell survival, and proliferation. It is expressed on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes but not on B or NK cells. Decreased T lymphocytes may be observed in certain autoimmune diseases.

Defects in CD3 genes are associated with CD3 immunodeficiency.
