Description:
The mT2.7 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by the myeloid lineage, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line.

Toll-like receptors (TLRs) are type I transmembrane proteins characterized by extracellular leucine-rich repeats and a cytoplasmic domain homologous to the IL-1 receptor. Among these, TLR2 and TLR4 act as pattern recognition receptors and signaling molecules responding to bacterial lipoproteins, playing key roles in innate immunity and inflammation.

TLR2 is expressed on the surface of cells and distinguishes different pathogens.

Applications Reported:

  • Suitable for flow cytometric analysis.

Applications Tested:

  • Tested by flow cytometric analysis of mouse splenocytes.
  • Recommended ≤ 0.5 µg per test (defined as staining a cell sample in 100 µL final volume).
  • Optimal performance should be determined empirically (cell number range: 10⁵–10⁸ cells/test).
  • Antibody should be carefully titrated for best results.

Target Information

TLR2 is a member of the Toll-like receptor family involved in pathogen recognition and activation of innate immunity.
TLR proteins signal through adaptor molecules such as MyD88 and TIRAP, activating kinases and transcription factors including PKC α/β and NF-κB.

TLR2 forms heterodimers with TLR1 or TLR6, allowing recognition of diverse bacterial and mycoplasma lipoproteins.
TLRs are evolutionarily conserved and exhibit distinct expression patterns.

TLR2 is most abundant in peripheral blood leukocytes, mediating host responses to Gram-positive bacteria and yeast via NF-κB activation.
It recognizes pathogen-associated molecular patterns (PAMPs) and induces cytokine production for effective immunity.

Human TLRs (TLR1–10) are expressed in various tissues such as tonsils, microglia, lymph nodes, and appendices.
CD14⁺ monocytes express the highest level of TLR2, followed by CD15⁺ granulocytes, CD19⁺ B-cells, and CD3⁺ T-cells.

Expression of TLR2 is modulated by immune response modifiers:

  • Up-regulated by: LPS, GM-CSF, IL-1, IL-10
  • Down-regulated by: IL-4, IFN-γ, TNF
