Product Specific Information
The 6C2 monoclonal antibody reacts with mouse Toll-like receptor 2 (TLR2). Mouse TLR2 is expressed by myeloid lineage cells, including macrophages and dendritic cells in splenocytes and the RAW264.7 cell line. The Toll family includes at least twelve members in humans and mice, characterized by an extracellular leucine-rich repeat domain and a cytoplasmic domain homologous to the type I IL-1 receptor. TLR2 and TLR4 are pattern recognition receptors involved in innate immunity and inflammation. TLR2 distinguishes pathogens at the cell surface.
Applications Reported:
The 6C2 antibody is reported for use in flow cytometric analysis and immunohistochemical staining of frozen tissue sections. For optimal flow cytometry results, use directly conjugated 6C2 (FITC, PE, or Biotin) with FcR blocking using anti-mouse CD16/CD32 (clone 93, Product #14-0161).
Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤1 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

0.2 µm post-manufacturing filtered.
Target Information
TLR2 is a member of the Toll-like receptor (TLR) family, essential for pathogen recognition and activation of innate immunity. TLR proteins signal through adaptor molecules such as MyD88 and TIRAP to activate kinases and transcription factors including PKCα/β and NF-κB. TLR2 forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins. TLR2 is abundantly expressed in peripheral blood leukocytes and mediates host responses to Gram-positive bacteria and yeast via NF-κB activation. Expression is highest in CD14⁺ monocytes, followed by granulocytes, B cells, and T cells. Cytokines such as LPS, GM-CSF, IL-1, and IL-10 upregulate TLR2, while IL-4, IFN-γ, and TNF downregulate it.
