RUNX2는 RUNX 전사인자 패밀리에 속하는 핵 단백질로, Runt DNA-binding domain을 포함합니다.
골막 및 연골 내 골 형성에 필수적이며, 골아세포 분화와 골격 형태 형성을 조절합니다. 또한 골격 관련 유전자 발현에 관여하는 핵산 및 조절 인자의 스캐폴드 역할을 수행합니다.
DNA에 단독으로 혹은 이합체 복합체의 서브유닛으로 결합할 수 있으며, 골아세포에서 TGFBR1 발현 증가에 중요한 역할을 합니다. DLX5 또는 관련因자와 협력하여 골아세포 특이적 유전자 발현을 활성화합니다.
RUNX2 변이는 쇄골두개 이형성증(Cleidocranial Dysplasia, CCD)과 같은 골 발달 장애를 유발할 수 있습니다.
