Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses.
TRAF proteins share a conserved region that enables binding to receptors and other TRAFs, activating distinct signaling cascades.
TRAF2 and TRAF3 interact with CD40, crucial for NF-κB activation in B lymphocytes.
TRAF2, together with TRAF6, is required for CD40 signaling in nonhematopoietic cells and also interacts with the lymphotoxin-beta receptor (LTβR) in association with TRAF3, cIAP1, and Smac.
