Product Specific Information

The 8A7 monoclonal antibody reacts with the human BAFF receptor (B lymphocyte stimulator receptor), expressed on B cells. BAFF is a B cell survival factor regulating CD21/35 and CD23 expression. Ligand-receptor interaction elevates CD21/35 and CD23 expression, while receptor blockade down-modulates expression.

Applications Reported:

• Flow cytometric analysis
• Immunoblotting (WB)
• Immunohistochemical staining
• Functional assays (use Functional Grade 8A7, Cat. 16-9117)

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤5 µL (0.4 µg)/test in 100 µL final volume. Cell number may range from 10⁵ to 10⁸ cells/test. Titrate for optimal performance.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used with NovaFluor conjugates for optimal performance.
  • Add 5 µL per sample to antibody cocktails/master mixes before adding to cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

Tumor necrosis factor receptor superfamily member 13C (TNFRSF13C), also known as BAFFR, is encoded by the TNFRSF13C gene on chromosome 22q13.1–q13.31. It encodes a 184 amino acid transmembrane protein (56% identical to mouse protein). BAFF enhances B-cell survival and regulates peripheral B-cell populations, playing a key role in B-cell development.
