Product Specific Information

The HI111 monoclonal antibody reacts with human CD11a, the 180 kDa integrin alphaL, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta2), serves as an important adhesion molecule involved in lymphocyte and granulocyte function. CD11a is expressed by all leukocytes.

Applications Reported:
The HI111 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.8 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL of CellBlox Plus per sample or per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 forms leukocyte function-associated antigen 1 (LFA-1), an αLβ2 integrin involved in leukocyte adhesion and signaling. CD11a interacts with ICAM-1, -2, and -3 and plays a critical role in immune cell adhesion, migration, differentiation, and activation. Dysregulation of CD11a is associated with leukocyte adhesion deficiency and inflammatory diseases. Therapeutic targeting of CD11a (e.g., with Efalizumab) can modulate immune cell migration and activity.
