Target Information

ATG5 (Autophagy Related 5) is a key component of the autophagy pathway and plays an important role in apoptotic processes. It is involved in mitochondrial quality control after oxidative damage, regulation of innate anti-viral immune response, lymphocyte development and proliferation, MHC II antigen presentation, and adipocyte differentiation.

Following conjugation to ATG12, the ATG5–ATG12 complex participates in autophagosome formation. ATG5 contributes to autophagic cell death by interacting with Fas-associated protein with death domain (FADD). The ATG5–ATG12 conjugate forms a cup-shaped isolation membrane that detaches before or after autophagosome completion. Additionally, APG5 expression occurs downstream of caspase activity during apoptosis and can interact with immune response proteins such as RIG-I and VISA, modulating interferon production and viral replication. Dysfunction of ATG5 has been associated with spinocerebellar ataxia.
