Target Information

EGF (Epidermal Growth Factor) binds to the EGF Receptor (EGFR), a 170 kDa protein kinase. Activation of EGFR initiates multiple cellular signaling pathways in response to environmental stimuli or ligand binding. EGFR forms homo- or heterodimers with other family members, each triggering distinct pathways via SH2-containing effector proteins.
EGF is widely expressed in kidney, cerebrum, prostate, and salivary glands, acting as a potent mitogenic factor. The phosphorylated receptor recruits adapter proteins such as GRB2, activating downstream cascades including RAS-RAF-MEK-ERK, PI3K-AKT, PLCγ-PKC, and STAT pathways.
Studies suggest EGF may also play a role in NF-κB signaling activation. Defects in the EGF gene are linked to hypomagnesemia type 4 and dysregulation has been associated with cancer progression.
