Product Specific Information

The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine. It is a neutralizing antibody that targets mouse IL-4 (also known as BCDF, BCGF-1, BSF-1). IL-4 is secreted primarily by activated Th2 and natural killer T cells, and promotes proliferation and differentiation of B cells. It also induces Ig isotype switching and upregulates surface MHC class II and CD23.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Validated using mouse Th2-polarized splenocytes with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Recommended use: ≤ 0.25 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test.
Antibody should be titrated for optimal performance.

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is intended for UV laser-equipped cytometers (355 nm).
Ensure instrument compatibility.

Staining Panel Considerations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples may be stored in:

• IC Fixation Buffer (Product # 00-8222-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)
  for up to 3 days at 4°C (dark) with minimal impact on brightness and FRET efficiency.

Target Information

Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells and binds to the interleukin 4 receptor. It also interacts with IL13, contributing to overlapping functions.
STAT6 mediates IL-4 signaling.
The IL-4 gene is part of a cytokine cluster on chromosome 5q, near IL13, IL5, IL3, and CSF2.
IL-4 δ2 is a known splice variant.
