Product Specific Information

Description:
The 4S.B3 monoclonal antibody reacts with human interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells. IFN-γ exhibits antiviral and antiparasitic activity and modulates immune responses via high-affinity receptors. Glycosylation does not affect its biological activity.

Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for intracellular staining of normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended usage: 5 µL (1.0 µg) per test in 100 µL final volume. Cell counts may vary from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is optimized for cytometers with a 355 nm UV laser. Ensure instrument compatibility before use.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal loss of brightness.

Target Information

IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor primarily produced by T-lymphocytes and NK cells in response to antigens and cytokines. It functions as a homodimer (~45 kDa), showing antiviral and antiproliferative effects, and induces MHC class I/II expression.
Human IFN-γ does not cross-react with mouse IFN-γ. Activity is regulated by IL-2, FGF basic, EGF (upregulation), and vitamin D3 or DMN (downregulation). Mutations are associated with aplastic anemia.
