Product Specific Information

Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFN gamma). Human IFN gamma is a 17 kDa factor produced by activated T and NK cells and functions as an antiviral and antiparasitic cytokine. It works synergistically with TNF alpha to inhibit cell proliferation and modulates immune responses via high-affinity receptors on various cell types. Glycosylation does not affect its biological activity.

Applications Reported:
Reported for use in intracellular flow cytometric analysis.

Applications Tested:
Validated for intracellular flow cytometry at ≤ 0.125 µg/test (100 µL final volume). Cell number per test should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal assay performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

IFN gamma (Interferon gamma, Type II interferon)
A macrophage activation factor and immune interferon produced mainly by T lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146-amino-acid subunits. Biological activity is species-specific; human IFN gamma does not cross-react with mouse. Functions include antiviral and antiproliferative activity, induction of MHC I/II, macrophage activation, and enhanced immunoglobulin secretion by B cells. It acts through the JAK-STAT pathway via IFN gamma receptor I and II. Expression is upregulated by IL-2, FGF basic, EGF, and downregulated by vitamin D3 or DMN. Mutations are associated with aplastic anemia.
