Product Specific Information
Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic activities. IFN-γ acts synergistically with cytokines such as TNF-α to inhibit cell proliferation and modulate immune responses across various cell types expressing IFN-γ receptors. Glycosylation does not affect its biological activity.
Applications Reported:
For intracellular flow cytometric analysis.
Applications Tested:
Pre-titrated and validated for intracellular flow cytometry. Use at 5 µL (0.125 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.
Light Sensitivity:
Protect vial and stained samples from light due to photo-induced oxidation.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor produced by T-lymphocytes and NK cells in response to antigens and cytokines. It is a dimeric glycoprotein (~45 kDa) composed of two 146 amino acid subunits. IFN-γ exhibits species-specific activity and does not cross-react between human and mouse. Functions include:
- Antiviral and antiproliferative activity
- Induction of MHC class I and II expression
- Macrophage activation
- Enhancement of immunoglobulin secretion by B lymphocytes
Activation occurs via IFN-γ receptor I/II and the JAK-STAT signaling pathway. Expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN. Mutations in the IFN-γ gene are associated with aplastic anemia.
