Target Information

DNA-binding protein SATB2 binds to DNA at the nuclear matrix- or scaffold-associated regions and recognizes the sugar-phosphate structure of double-stranded DNA.
SATB2 acts as a transcription factor that regulates gene expression by binding to matrix attachment regions (MARs) and inducing chromatin-loop remodeling. It also serves as a docking site for chromatin remodeling enzymes, recruiting corepressors (HDACs) or coactivators (HATs) directly to promoters and enhancers.
SATB2 is required for the initiation of upper-layer neuron (UL1) genetic programs and the inactivation of deep-layer neuron (DL) and UL2 genes, likely through modulation of BCL11B expression. It functions as a repressor of Ctip2 and a key determinant of corticocortical connections in the developing cerebral cortex. Mutations in SATB2 are associated with cleft palate isolated (CPI).
