Semaphorins are the largest family of axon guidance cues, divided into 8 classes (classes 3–7 found in vertebrates).
- Class 3 Semaphorins: Secreted proteins
- Classes 4–6: Transmembrane proteins
- Class 7: Membrane-associated via GPI linkage
They contain a conserved ~400 amino acid sema domain and function as collapsing factors and mediators of axon repulsion, while also acting as context-dependent chemoattractants.
Semaphorins are involved in cardiovascular development and immune cell antigen presentation regulation.
Receptors mediating semaphorin signaling include Neuropilin and Plexin families.
