Target Information
Methylation of DNA at cytosine residues in mammalian cells is a heritable, epigenetic modification essential for proper gene expression, genomic imprinting, and development. Three families of mammalian DNA methyltransferases have been identified: DNMT1, DNMT2, and DNMT3.
- DNMT1: Maintenance methyltransferase, active during DNA replication.
- DNMT3A and DNMT3B: De novo methyltransferases, highly expressed in embryonic stem cells.
- DNMT2: Low expression in adult tissues, minimal impact on DNA methylation.
- DNMT3L: Catalytically inactive regulatory factor for DNMT3A and DNMT3B, expressed in embryonic stem cells, testis, ovaries, and thymus. Forms heterotetrameric complexes with DNMT3A or DNMT3B and contains an N-terminal ADD domain and a C-terminal methyltransferase-like domain.
