Post Synaptic Density 95 kDa (PSD-95), also known as synapse associated protein 90 kDa (SAP90), is a membrane-associated protein found in the postsynaptic density of forebrain neurons and certain presynaptic structures in the cerebellum.
PSD-95 contains three PDZ domains, an SH3 domain, and a GuK domain. It plays a key role in clustering NMDA receptors and Shaker-type potassium channels at the synaptic membrane in central nervous system neurons.

Two principal modes of interaction exist between PSD-95 and other proteins:

1. NMDA receptors and Shaker-type potassium channels share a C-terminal T/SXV motif that binds to PSD-95 PDZ domains.
2. Neuronal nitric oxide synthase (nNOS), lacking the T/SXV motif but containing its own PDZ domain, interacts with PSD-95 through PDZ-PDZ interaction.
