Target Information

Apoptosis is a key process for maintaining normal tissue homeostasis. Activation of the Fas receptor by its ligand triggers formation of a Fas-associated death domain (FADD)-containing signaling complex and activation of caspase-8, which subsequently activates downstream caspases such as caspase-3 and -7.
Overexpression of Toso, a novel regulator of Fas-induced apoptosis in lymphoid cells, renders Jurkat and transgenic mouse cells resistant to Fas-induced apoptosis but not to TRAIL-induced apoptosis. Toso associates with FADD, suggesting it disrupts formation of the death-inducing signaling complex.
