Target Information
Crystallins are divided into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous class forms the major proteins of vertebrate eye lenses, maintaining lens transparency and refractive index. Lens central fiber cells lose nuclei during development, so crystallins are synthesized once and retained throughout life, making them highly stable.
Mammalian crystallins are categorized into alpha, beta, and gamma families, with beta and gamma forming a superfamily. Alpha and beta families are further divided into acidic and basic subgroups. Crystallins possess seven protein regions: four homologous motifs, a connecting peptide, and N- and C-terminal extensions.
Gamma-crystallins are monomeric, highly symmetrical proteins typically lacking connecting peptides and terminal extensions, and are differentially regulated after early development. The gene encoding Beta crystallin S was initially classified as beta-crystallin but shows greater similarity to gamma-crystallins and encodes the predominant gamma-crystallin in adult eye lens tissue. Mutations or aging-related changes in gamma-crystallins are associated with cataract formation.
