Target Information
TRIP12 is an E3 ubiquitin-protein ligase involved in the ubiquitin fusion degradation (UFD) pathway and regulation of DNA repair. It mediates ubiquitination of proteins at their N-terminus, independent of lysine residues. In normal cells, TRIP12 targets isoform p19ARF/ARF of CDKN2A for degradation, regulating p53 activation. In cancer cells, compartmental separation prevents this degradation. TRIP12 also ubiquitinates NAE1 and SMARCE1, leading to their degradation. Its activity is modulated by interactions with proteins such as MYC, TRADD, and SMARCC1. TRIP12 acts as a suppressor of RNF168, preventing excessive accumulation of ubiquitinated histones at DNA damage sites.
