Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and are involved in protein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit, with approximately 75% protein to rRNA composition, unlike prokaryotic ribosomes where this ratio is reversed. Mammalian mitoribosomes lack a 5S rRNA found in prokaryotic ribosomes. The proteins comprising the mitoribosome vary significantly across species, making sequence homology identification difficult. The MRPL43 gene encodes a 39S subunit protein and overlaps with the SEMA4G gene at map location 10q24.31, transcribed in opposite directions. Multiple transcript variants encoding at least four protein isoforms have been identified.
