Target Information
NUAK1 (ARK5) is a serine/threonine kinase involved in multiple cellular functions. Overexpression of this kinase is associated with tumor invasion and metastasis, particularly in colorectal neoplasms. It regulates cellular senescence and ploidy through phosphorylation of Ser-464 on LATS1, influencing its stability. Additionally, ARK5 controls cell adhesion by modulating the myosin protein phosphatase 1 (PP1) complex via phosphorylation of PPP1R12A, leading to altered myosin MLC2 dephosphorylation. It participates in DNA damage response by phosphorylating p53/TP53 at Ser-15 and Ser-392 and contributes to transcriptional activation at the CDKN1A/WAF1 promoter. ARK5 may act as a tumor malignancy-associated factor under AKT1 regulation, suppressing Fas-induced apoptosis through CASP6 phosphorylation. It also plays a role in UV-induced DNA damage response by phosphorylating CDKN1A in association with STK11, aiding its degradation for optimal DNA repair.
