Target Information
The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain. AR acts as a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates to the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
AR signaling is essential for the development and function of male reproductive organs such as the prostate and epididymis, and also plays roles in muscle, hair follicles, and brain. As a phosphoprotein, AR regulates MAP kinase pathways, and its phosphorylation state is influenced by MEK1/2 pathway activity. Dysregulation of AR signaling has been linked to diseases including prostate cancer, Kennedy’s disease, male infertility, and complete androgen insensitivity syndrome (CAIS).
