Target Information
RUSC1, also known as NESCA, shares with the related protein RUSC2 a common domain structure consisting of RUN, leucine zipper, and SH3 domains, with over 30% amino acid identity.
RUSC1 acts as an adaptor protein that binds to the TrkA receptor and is essential for NGF-induced neurite growth in PC12 cells. It interacts with IKK-γ (the regulatory subunit of the IKK complex) and TRAF6, influencing NF-κB activation pathways such as TNF-R and TLR signaling.
Overexpression of RUSC1 inhibits TRAF6-mediated polyubiquitination of IKK-γ, suggesting RUSC1 plays a regulatory role in NF-κB signaling.
