Human TRAIL (TNF-Related Apoptosis Inducing Ligand), also called Apo2 Ligand, is a cytotoxic protein that activates rapid apoptosis in tumor cells but not in normal cells.
Human TRAIL/Apo2 Ligand is a 19.6 kDa protein comprising the full-length TNF-like extracellular domain of TRAIL.

Target Information

TRAIL is a cytokine belonging to the tumor necrosis factor (TNF) ligand family. It preferentially induces apoptosis in transformed and tumor cells, while sparing normal cells despite its expression in most tissues.
TRAIL binds to several TNF receptor superfamily members, including:

  • TNFRSF10A/TRAILR1
  • TNFRSF10B/TRAILR2
  • TNFRSF10C/TRAILR3
  • TNFRSF10D/TRAILR4
  • Possibly TNFRSF11B/OPG

Its activity may be modulated by decoy receptors (TRAILR3, TRAILR4, OPG) that do not induce apoptosis. Binding of TRAIL to its receptors triggers activation of MAPK8/JNK, caspase 8, and caspase 3.
Receptors include two death domain-containing receptors (DR4, DR5) and two decoy receptors (DcR1, DcR2) lacking intracellular signaling domains.
TRAIL is a type II membrane protein expressed in various human tissues and, like TNF and Fas ligand, induces apoptosis and NF-κB activation.
