Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis, implicated in retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis of apoptotic cells. Ligand binding induces autophosphorylation of MERTK, enabling downstream signaling through GRB2 or PLCG2, and phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance, platelet aggregation, cytoskeleton reorganization, and inhibition of TLR-mediated innate immune response by activating STAT1 and inducing SOCS1/SOCS3 production.
