Target Information
Claudin-18은 tight junction 단백질로, 폐(lung) 및 위(stomach) 특이적 isoform이 존재하며 claudin-18 유전자의 alternative splicing에 의해 생성됩니다. 폐 특이적 형태는 T/EBP/NKX2.1 전사인자에 의해 조절되는 하위 표적 유전자입니다.
마우스에서 C-말단 세포질 도메인이 결여된 splice variant가 존재하나, 인간에서는 확인되지 않았습니다. 인간 Claudin-18은 마우스 단백질과 88%의 아미노산 서열 동일성을 보입니다.
면역조직화학 연구에서는 Claudin-18이 폐 및 위 상피세포의 세포막에 완전히 국한되어 있으며, 전자현미경 관찰에서는 세포 간 경계 부위에 집중되어 있음이 확인되었습니다. 이러한 특징은 Claudin-18이 폐 및 위의 tight junction 구조와 기능에 중요한 역할을 할 가능성을 시사합니다. 또한 Claudin-18의 발현은 내이(inner ear)에서도 보고되었습니다.
