Target Information

Isoform 1 of Clusterin prevents stress-induced aggregation of blood plasma proteins and inhibits formation of amyloid fibrils by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro). It maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70. Clusterin does not refold proteins by itself but facilitates internalization and degradation of chaperone-client complexes. Intracellular isoforms interact with ubiquitin and SCF E3 ligase complexes, promoting proteasomal degradation of target proteins such as COMMD1 and IKBKB, and modulating NF-kappa-B activity. Nuclear isoforms promote apoptosis, while mitochondrial isoforms inhibit BAX-dependent cytochrome c release and apoptosis. Plays a role in regulating cell proliferation.
