Target Information

The leptin (OB-R) receptor is a member of the cytokine receptor superfamily that plays an important role in mammalian body weight homeostasis and energy balance. Activation of OB-R by leptin triggers downstream signaling through the JAK/STAT pathway. Leptin can induce homo-dimerization of OB-R extracellular and intracellular domains, leading to signal transduction. Multiple isoforms of OB-R with different signaling capacities have been identified. Expression of the long form of OB-R is found in the choroid plexus and hypothalamus, key regions for satiety and energy regulation. Mutations in OB-R (db) or leptin (ob) genes are linked to early-onset obesity and associated metabolic abnormalities, such as hyperinsulinemia, insulin resistance, and altered CNS activity.
