Product Specific Information

PA3-026 detects calcineurin inhibitor (cain) from human and rat tissues.
It has been successfully used in Western blot, immunoprecipitation, immunofluorescence, and ELISA procedures.
By Western blot, this antibody detects an approximately 240 kDa protein representing cain from rat liver extract.
Immunoprecipitation of cain from rat brain tissues shows an interaction between cain and amphiphysin 1, adaptin, and dynamin 1.

The immunizing peptide is a His-tagged fusion protein corresponding to amino acid residues 1881–2173 from rat cain.


Target Information

Calcineurin binding protein (Cabin-1) and its rat homolog, Cain, are nuclear phosphoproteins that regulate the serine/threonine phosphatase activity of calcineurin and influence T cell signaling and apoptosis.
Calcineurin is essential for cytokine transcriptional activation and the activation of transcription factors such as NFAT, NFκB, and AP-1 in T cell receptor (TCR)-mediated signaling.
TCR activation triggers PKC-induced hyperphosphorylation of Cabin-1, enhancing its binding affinity to calcineurin and inhibiting calcineurin activity, thereby attenuating TCR-mediated signaling.
Cabin-1 also associates with MEF-2 transcription factors, inhibiting MEF-2-mediated gene transcription and apoptosis signaling in T cells.
