Product Specific Information

The monoclonal antibody ME20.4 recognizes CD271, a 75 kDa type I transmembrane protein belonging to the low-affinity neurotrophin receptor and tumor necrosis factor receptor superfamily. CD271 is expressed by neurons, Schwann cells, melanocytes, mesenchymal stem/stromal cells, neural crest stem cells, astrocytes, oligodendrocytes, and certain lymphoid cells including follicular dendritic cells.

CD271 binds to NGF, BDNF, NTF3, and NTF4, and contains a death domain that induces apoptosis and regulates cell growth and proliferation through interactions with TRAF2, TRAF4, TRAF6, PTPN13, and RANBP9.

The ME20.4 antibody recognizes CD271 from human, non-human primate, mouse, rat, sheep, rabbit, cat, dog, and pig.

Applications Reported: Flow cytometric analysis, western blotting, microscopy, immunohistochemical staining, and immunocytochemistry.

Applications Tested:

• Flow cytometric analysis of SK-N-SH cells (≤0.25 µg/test; 100 µL final volume; 10^5–10^8 cells/test)
• Immunohistochemistry of formalin-fixed paraffin-embedded human tissue using high pH antigen retrieval (≤5 µg/mL)
• Antibody titration recommended for optimal assay performance.

Target Information

NGF is essential for developing sympathetic neurons during target competition in vivo. Antibodies specific to NGFR p75 are valuable tools for detecting and characterizing NGFR p75, aiding research on neural development, plasticity, and repair.
