The eBio18F10 antibody reacts with mouse IL-17F, a 37 kD homodimeric cytokine belonging to the IL-17 family and a signature Th17 marker. IL-17F and IL-17A share strong sequence homology and are co-regulated during Th17 differentiation. Both are expressed in activated human CD4+ T cells and are associated with autoimmune and inflammatory diseases such as rheumatoid arthritis, multiple sclerosis, psoriasis, inflammatory bowel disease, and asthma.
IL-17F induces expression of IL-6, IL-8, GRO-alpha, ENA-78, TGF-beta, MCP-1, G-CSF, GM-CSF, and ICAM-1 in airway epithelial and endothelial cells. It forms both homodimers and IL-17A/F heterodimers, which vary in biological potency.
Applications Reported: Useful for intracellular staining and flow cytometric analysis.
Applications Tested: Tested on in vitro-differentiated mouse Th17 cells and EL-4 cell line. Recommended ≤ 0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.
Excitation/Emission: 488 nm / 519 nm (Blue Laser).
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
IL-17F (Interleukin 17F, CTLA-8) is a cytokine produced by activated T cells, mast cells, and basophils. It induces cytokine and chemokine expression in epithelial, endothelial, and fibroblast cells, contributing to allergic and inflammatory responses. IL-17F can form homodimers or heterodimers with IL-17A, acting via the IL-17R receptor to promote monocyte and neutrophil recruitment.
IL-17F is implicated in chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis. It is regulated by TGF-beta and IL-23 during Th17 differentiation and inhibits angiogenesis while inducing IL-2, TGFB, and MCP-1 production in endothelial cells.
