Product Specific Information

The HB15e monoclonal antibody reacts with human CD83, a 45 kDa transmembrane glycoprotein belonging to the Ig superfamily. CD83 is expressed on cultured dendritic cells, interdigitating and follicular dendritic cells, circulating dendritic cells, and some proliferating lymphocytes. It serves as a useful marker for mature human blood dendritic cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of stimulated normal human peripheral blood cells.
Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited by the Red laser (633–647 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD83 is a 40–45 kDa glycoprotein expressed on peripheral blood dendritic cells. Peripheral lymphocytes can express low levels of CD83 after stimulation with Con A or PHA. In immunohistology, CD83 is strongly expressed by interfollicular interdigitating reticulum cells and weakly by germinal center cells, also found on Langerhans cells in skin.

The CD83 antigen is a 186-amino-acid single-chain glycoprotein, a member of the immunoglobulin superfamily with an extracellular V-type Ig-like domain, transmembrane region, and short cytoplasmic tail. It undergoes extensive post-translational glycosylation, doubling its molecular weight compared to the core protein.

CD83⁺ cells coexpress high levels of MHC class II molecules and various lineage markers (CD2, CD5, CD40, CD78, CD13, CD33, CD36). Diseases associated with CD83 dysfunction include plague and Rift Valley Fever.
