Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells and binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
CTLA-4 and CD28 deliver opposing signals: CD28 acts as a costimulator, while CTLA-4 inhibits T cell activation by restricting IL-2 secretion and proliferation.

A large portion of CTLA-4 is localized intracellularly due to ER retention motifs.
This antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitor activity.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).

For complete detection, assess both surface and intracellular CTLA-4 expression.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed mainly on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28, inhibiting T cell activation. Intracellular CTLA4 is also present in regulatory T cells and contributes to their suppressive function.
