Target Information
Cas9 is an RNA-guided DNA endonuclease enzyme associated with the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) adaptive immunity system in bacteria and genetic engineering. The system depends on RNA to target the nuclease to a desired DNA sequence, enabling indel mutations, sequence replacements, insertions, and genomic rearrangements at specific loci.
Cas9 can also be used to upregulate endogenous genes or modify histone and DNA methylation patterns.
Cas9 variants include Streptococcus pyogenes (SpCas9) and Staphylococcus aureus (SaCas9). SaCas9 is smaller (1053 AA vs. 1368 AA), highly efficient, and specific, recognizing NNGRRT PAM sequences. It is ideal for all-in-one delivery of Cas9 and multiple gRNA cassettes using adeno-associated vectors.
