Product Specific Information

This TM-724 monoclonal antibody reacts with human Autoimmune Regulator (AIRE), a 58 kDa transcription factor highly expressed in the perinuclear region of mature medullary thymic epithelial cells (mTECs). AIRE mRNA is also found in lymph nodes, fetal liver, intestines, dendritic cells, epithelial cells, and PBMCs.
AIRE is involved in thymocyte development and regulation of tissue-specific antigen expression during negative selection. It also influences chemokine expression and antigen presentation. Three isoforms (AIRE-1, AIRE-2, AIRE-3) have been reported, and this antibody is predicted to recognize all three.
Mutations in AIRE cause autoimmune-polyendocrinopathy-candidiasis-ectodermal-dystrophy (APECED) and autoimmune polyglandular syndrome type 1 (APS-1).

Applications Reported: Suitable for IHC (FFPE), microscopy, immunohistochemical staining, and immunocytochemistry.
Applications Tested: Validated on FFPE human thymus at ≤10 µg/mL using low pH antigen retrieval. Optimize dilution for specific assays.
Filter Recommendation: Use a filter capturing 570 nm emission (e.g., Excitation 545/25, 565LP, Emission 605/70). Alexa Fluor® 555 or TRITC filters are acceptable.
Excitation/Emission: 555 nm / 570 nm.


Target Information

AIRE is a transcriptional regulator protein that binds DNA as a dimer and tetramer. It contains an HSR domain, two PHD-type zinc fingers, and a SAND domain. AIRE interacts with coactivators CBP and CREBBP and can form nuclear bodies.
It plays a key role in immune regulation and exhibits E3 ubiquitin ligase activity. Mutations in AIRE lead to the rare autosomal-recessive disease APECED.
