Target Information

The Munc-18 interacting protein (Mint) family comprises evolutionarily conserved adaptor proteins involved in membrane transport and organization. Mammals express three isoforms—Mint1, Mint2, and Mint3. The C-terminal halves of these proteins are highly conserved and contain phosphotyrosine-binding (PTB) and PSD-95/DLG-A/ZO-1 (PDZ) domains, which serve as protein interaction modules.

Mint1 and Mint2 are expressed primarily in the brain and interact with Munc18, a critical component of synaptic vesicle fusion. Mint3, however, is ubiquitously expressed, with the lowest levels in the brain and testis, and does not interact with Munc18. Mint3 associates with the Alzheimer’s disease-related amyloid precursor protein (APP) through its PTB and PDZ domains, linking APP to transport machinery components that regulate its trafficking, endocytosis, and metabolism. Abnormal APP metabolism is implicated in early-onset Alzheimer’s disease.
