Target Information
Casein kinase I (CKI) and casein kinase II (CKII) are families of serine/threonine protein kinases found in all eukaryotes. CKI family members, including CKI alpha, gamma, epsilon, and delta, are involved in regulating cytoplasmic and nuclear processes such as DNA replication and repair.
CKII typically exists as a tetrameric complex (α₂β₂ or αα′β₂), where the alpha catalytic subunit is stimulated by the beta regulatory subunit, which undergoes autophosphorylation. CKII activity is abundant in the cytosol and nucleus of proliferating and differentiating cells and is known to phosphorylate over 100 substrates, including nuclear oncoproteins, transcription factors, and DNA metabolism enzymes.
