Target Information

Ubiquitin-mediated proteolysis requires the transfer of ubiquitin (Ub) to lysine groups on selected cellular proteins, leading to their degradation by the 26S proteasome. Ub-fusions are cleaved by Ub-specific processing proteases (UBps) or by the Ub-fusion degradation (UFD) pathway.
The UFD1 protein, originally identified in S. cerevisiae, has a human homolog (UFD1L) that modulates degradation of Ub-fusions with substitutions at the Gly76 residue. Mutations in the UFD1 gene are linked to CATCH22 syndrome, which involves cardiac defects, cleft palate, and hypocalcemia, suggesting a role in developmental processes.
