The tumor necrosis factor (TNF) receptor superfamily consists of type I integral membrane glycoproteins with cystine-rich extracellular domains. Members include TNF-RI, TNF-RII, and CD40. Ligands for these receptors can be secreted proteins (e.g., TNF) or membrane proteins (e.g., CD40L). Activation of TNF-R or CD40 induces nuclear translocation of NFκB. TRAF (TNF receptor-associated factor) family members mediate this process. Four members—TRAF1, TRAF2, TRAF3, and TRAF4—have been identified. TRAF4, also known as CART1, is specifically expressed in breast carcinomas and localized to the nucleus in such tissues.
