Target Information
NANOG (Nanog homeobox) is a divergent homeodomain protein that regulates pluripotency and differentiation of undifferentiated embryonic stem cells. NANOG mRNA is expressed in pluripotent mouse and human cell lines, but absent from differentiated cells. Human NANOG protein shares 52% overall amino acid identity with the mouse protein and 85% identity in the homeodomain.
Human NANOG maps to gene locus 12p13.31, while mouse NANOG maps to 6 F2. Murine embryonic NANOG expression is detected in the inner cell mass of the blastocyst. High levels of NANOG expression are found in undifferentiated N-Tera embryonal carcinoma cells.
NANOG acts as a transcription regulator involved in inner cell mass and embryonic stem (ES) cell proliferation and self-renewal. Its role in embryonic development suggests potential applications in creating stem cells for cell replacement therapies in degenerative disease treatment.
Artificial stem cells, termed induced pluripotent stem (iPS) cells, can be generated by expressing POU5F1 (Oct-4), Sox2, Klf4, and Lin28 along with c-Myc in mouse fibroblasts. Studies show iPS cells can be created using plasmids expressing NANOG, Sox2, Klf4, and c-Myc, eliminating viral introduction and improving safety for regenerative medicine.
Overexpression of NANOG promotes S phase entry and cell proliferation. Diseases associated with NANOG dysfunction include teratocarcinoma and germ cell/embryonal cancer.
