The L243 monoclonal antibody recognizes human major histocompatibility complex (MHC) class II, HLA-DR, expressed on antigen-presenting cells such as B cells, monocytes, macrophages, dendritic cells, and activated T cells.
HLA-DR is a heterodimeric transmembrane protein composed of alpha and beta subunits, crucial for peptide presentation to CD4+ T lymphocytes.
The L243 antibody binds a distinct epitope compared to LN3 and does not cross-block.

Target Information

HLA-DR is a transmembrane glycoprotein consisting of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The DRA gene contains 5 exons encoding leader peptide, extracellular domains, transmembrane region, and cytoplasmic tail.
DRA is non-polymorphic, while DRB contains polymorphisms determining peptide binding specificity.
Hundreds of DRB1 alleles are known, routinely typed for bone marrow and kidney transplantation.
HLA-DR is expressed mainly on antigen-presenting cells (B lymphocytes, monocytes, macrophages, thymic epithelial cells, activated T lymphocytes).
MHC class II molecules (DR, DQ, DP) present processed peptides to T-helper cells, initiating immune responses.
HLA and MHC antibodies are essential in Immunopeptidomics, aiding neoantigen identification via LC-MS/MS.
