Description:
The monoclonal antibody MH5A reacts with a homolog of PD-1 (PD-1H), also known as VISTA. PD-1H is a 309 amino acid type I transmembrane protein belonging to the CD28 superfamily. This receptor is expressed on nearly all hematopoietic cells except B cells and negatively regulates T cell responses by suppressing early TCR activation, inhibiting proliferation, and reducing TH1 cytokine production.

The MH5A antibody has been reported to inhibit T cell infiltration in graft-versus-host disease mouse models.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤ 0.25 µg/test (in 100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal assay performance.

Use in a sterile environment.

Target Information

VISTA (V-domain Ig containing suppressor of T-cell activation), also known as B7-H5 or PD-1H, is encoded by the VSIR gene. Although VISTA shares sequence homology with PD-L1, it exhibits a distinct expression pattern. In both mouse and human, VISTA is predominantly expressed on hematopoietic cells, with high levels on myeloid cells and lower levels on T cells, while absent on B cells.

VISTA expressed by antigen-presenting cells directly suppresses proliferation and cytokine production of CD4 and CD8 T cells, making it a promising target for cancer immunotherapy. Reported ligands include CD28H, VSIG-3, and VSIG-8.
