Target Information

SIRP alpha (CD172a, signal-regulatory protein alpha) is a receptor-type transmembrane glycoprotein expressed on myeloid cells including granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells.
It acts as a substrate for several activated tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase-coupled signaling pathways.
Binding of SIRP alpha to CD47 induces phosphorylation of ITIMs and recruitment of phosphatases SHP-1 and SHP-2, regulating processes such as inhibition of macrophage phagocytosis and activation of T cells and dendritic cells.
SIRP alpha influences responses to serum, growth factors, insulin, oncogenes, and cell adhesion, playing a role in physiological and pathological processes.
Cancer cells highly express CD47, which activates SIRP alpha and inhibits macrophage-mediated destruction of tumor cells.
